I understand that you should remove the air cleaners and hold the throttle wide open when doing a compression test but how many kick on the kick-starter should I give it? Three or four? Or how many kicks?

Removing the air cleaners isn't required. Give it as many kicks as you like, after 2 it shouldn't make any difference.
